Botanical name: Lespedeza seiboldii.
Family Papilionaceae Fabaceae
Uses: Nephritis and nephrosclerosis.
Description: Fresh aerial parts of flowering specimens of Lespedeza thunbergii (DC) Nakai.
The long pendulous branches of this up to 2 m high shrub or half-shrub are grooved and when juvenile hairy. The periolate leaves are up to 4 am long, alternate, trifoliate, with the terminal leaflet clearly periolate, and have two frail subulate stipules. The leaflets are 3-5 cm long, 1.5 - 2.0 cm wide elliptical elongated, with margins entire. They terminate in a short point. The upper surface of the lamina is glabrous, the underside covered with procumbent hairs from the axils of subulate bracts in racemes up to 25 cm long that combine to form spikes up to 80 cm long, with leaves present throughout. The pedicel is up to 7 mm long; a short distance below the gamosepalous 4-dentate calyx are two subulate bracts. The standard of the flower is erect or reflexed, obovate with short claws. The keel is at a downward angle; it is only slightly shorter than the standard and almost twice the length of the two clawed wings. The nine stamens are fused in a channel and fise at the apex; the tenth stamen covers the channel. The superior ovary contains a single ovule.
History and authority: It is an official drug of the German Homoeopathic Pharmacopoeia.
Part used: Aerial parts with flowers.
Preparation: (a)
Mother Tincture ø Drug strength 1/10
Lespedeza thunbergii
Moist magma containing solids 100 g
Plant moisture/Purified Water 300 ml
Strong Alcohol 730 ml

To make one litre of the Mother Tincture.
Potencies: 2x with 70% Alcohol, 3x and higher with Dispensing Alcohol.
Prescribed dose: 1x and higher
Storage: To be protected from light.
Characteristics: The mother tincture is a yellowy brown liquid with aromatic odour.
Identification of the mother tincture: a) To 1 ml of the mother tincture add 50 mg of magnesium filings and 1 ml of hydrochloric acid 1. A red colour is produced.
b) To 1 ml of the mother tincture add 2 ml of dilute hydrochloric acid. A precipitate is produced immediately.
c) To 1 ml of the mother tincture add 5 ml of water and 0.1 of lead (ii) acetate solution. A voluminous precipitate is produced.
d) Evaporate 0.2 ml of the mother tincture and add 0.5 ml of a 1 per cent solution (W/v) of vanillin in hydrochloric acid. An orange red colour is produced.
e) Chromatography. Use thin-layer chromatography in a layer of silica gel.
Test solution: The mother tincture.
Control solution: Dissolve 5 mg of hyperoside, 5 mg of caffeic acid and 10 mg of rutin in 10 ml of methanol.
Apply separately 40 µl of the test solution and 10 µl of the control solution. The mobile phase is a mixture of 80 parts by volume of ethyl acetate. 10 parts by volume of anhydrous formic acid and 10 parts by volume of water. Allow the solvent front to rise 15 cm above the line of application. Following evaporation of the mobile phase, spray the chromatograms with a 1 per cent solution (w/v) of polythylene glycol 400 in methanol. Evaluate under ultra-violet light (365 nm).
The Chromatogram of the control solution has the orange yellow fluorescent rutin spot in the lower third, the orange yellow fluorescent hyperoside spot in the lower part of the middle third and the bluish green fluorescent caffeic acid spot in the upper part of the upper third.
The chromatogram of the test solution has the following yellow green or yellow orange fluorescent spots: one each immediately below and above the rutin standard, one or two each below and above the hyperoside standard and one immediately above the caffeic acid standard.
Assay for purity: Relative density (Eur. P.): 0.895 - 0.915
Dry residue (German P.): Not less than 2.0 per cent.
Storage: Protected from light.
